Description 5-TAMRA-Aβ(1-40) is the 5-carboxytetramethylrhodamine conjugate of Aβ1-40 (Ex/Em ~557/570 nm, orange-red). Paired with 5-FAM/FITC-Aβ it forms a FRET dual-color system to report interpeptide proximity, oligomerization and fibrillation; the long wavelength also reduces tissue-autofluorescence background for amyloid imaging and inhibitor screening.

Physicochemical Solubility & Storage Parameters

Product Code Appearance Long-term Storage Short-term Storage Solution Stability Solid Powder Stability
PEP-TAMRAAB40-01 Pink to red powder (TAMRA rhodamine pigment) Store at -20C, sealed, desiccated, LIGHT-PROTECTED (amber tube/aluminum foil), ~2 years; strongly recommend aliquoting to avoid repeated freeze-thaw. The powder must be light- and moisture-protected. Aβ(1-40) is highly hydrophobic and aggregation-prone-HFIP pre-treat and aliquot immediately upon receipt; contains oxidation-prone Met-flush with argon/nitrogen. 2-8C, light-protected, for ~2 weeks; working aliquots at -80C, light-protected (after HFIP pre-treatment); avoid room-temperature exposure. Ship on dry ice, protected from light. Aβ(1-40) is strongly hydrophobic (C-terminal IIGLMVGGVV region); poor water solubility and spontaneous self-assembly into oligomers/fibrils. MUST follow the standard protocol: (1) reconstitute powder in hexafluoroisopropanol (HFIP) to 1 mM; sonicate/vortex to disaggregate pre-existing oligomers; (2) incubate at RT for 2-4 h to fully dissociate aggregates; (3) aliquot and blow dry HFIP with nitrogen/argon; (4) store dried films at -80C; before use, freshly reconstitute in DMSO or neutral buffer (pH 7.4 PBS/HEPES); (5) prepare fresh and avoid repeated freeze-thaw. PROTECT TAMRA FLUORESCENCE FROM LIGHT throughout (aluminum foil). Commonly mixed with FAM-Aβ(1-40) at defined ratios for FRET co-aggregation; Met35 is oxidation-prone and oxidized Aβ has altered aggregation behavior-control oxygen. solid stable ~2 years at -20C, sealed, desiccated, light-protected; contains TAMRA (light-sensitive) and Met (oxidizable)-store light-protected, dry, under inert gas. HFIP pre-treatment and aliquoting upon receipt is recommended to destroy pre-aggregates.

Functional Description

Product Code Frontend Short Summary Detailed Biological Function Description Applicable Research Areas (comma-separated) Targets / Pathways
PEP-TAMRAAB40-01 N-terminally 5-TAMRA orange-red fluorescent-labeled Aβ(1-40) (λex/em≈544/570 nm) for Alzheimer's disease Aβ aggregation FRET/ThT co-aggregation, dual-label aggregation kinetics paired with FAM-Aβ, and fluorescent tracing; highly hydrophobic and aggregation-prone-requires HFIP pre-treatment, light-protected aliquoting, and -80C storage. Amyloid-β (Aβ) is the primary protein component of senile plaques in Alzheimer's disease (AD) brain, generated by sequential cleavage of the β-amyloid precursor protein (APP) by β-secretase (BACE1) and γ-secretase. The major species are Aβ(1-40) (~90%) and Aβ(1-42) (~10%, but aggregates faster and is more neurotoxic). Aβ(1-40) is a 40-mer whose N-terminal region (residues 1-28) is relatively hydrophilic and charged, while the central hydrophobic core (LVFFAED, residues 17-24) and the C-terminal highly hydrophobic region (KGA IIGLMVGGVV, residues 30-40) jointly drive self-assembly from random-coil monomers → α-helical intermediates → β-sheet oligomers → mature fibrils; soluble oligomers are considered the main neurotoxic effector in AD, causing synaptic damage, mitochondrial dysfunction, oxidative stress, and microglial activation. N-terminally 5-TAMRA (5-carboxytetramethylrhodamine)-labeled Aβ(1-40) retains full aggregation properties and neurotoxicity while providing an orange-red fluorescent tracing channel (λex≈544 nm, λem≈570 nm), widely used for: (1) FRET co-aggregation assays PAIRING with FAM-Aβ(1-40)-TAMRA serves as the FRET acceptor and FAM as the donor; mixed at defined ratios, donor quenching/acceptor sensitization reports Aβ monomer-oligomer-fibril aggregation kinetics and nucleation-elongation curves in real time; (2) ThT fluorescence co-aggregation, fluorescence polarization (FP) or autocorrelation to determine oligomerization extent and aggregation-inhibitor IC50; (3) red-fluorescence tracing of Aβ uptake, endocytosis and toxicity in cells/organoids; (4) comparing aggregation kinetics with Aβ(1-42) or mutants (E22Q Dutch, E22G Arctic, etc.); (5) in vitro screening of AD therapeutic candidates (aggregation inhibitors, anti-Aβ antibodies lecanemab/donanemab). KEY NOTES: Aβ(1-40) is extremely hydrophobic; pre-formed oligomers may exist in the powder and must be dissociated with HFIP, blown dry under nitrogen, and aliquoted at -80C; reconstitute freshly in DMSO/neutral buffer before use and avoid repeated freeze-thaw; the TAMRA label is light-sensitive-operate under reduced light throughout; Met35 is oxidation-prone and oxidized Aβ has altered aggregation behavior-strictly control oxygen. Appearance: pink-to-red powder (rhodamine pigment). Alzheimer's disease (AD), Aβ aggregation kinetics/fibrils, FAM-TAMRA FRET co-aggregation, ThT fluorescence co-aggregation, AD drug/aggregation inhibitor screening, neurotoxicity/synaptic damage, fluorescent tracing of cellular uptake, oligomer FRET/FP analysis Aβ(1-40) self-aggregation (monomer → oligomer → fibril); TAMRA as FRET acceptor (paired with FAM donor); TAMRA fluorescence channel λex/em≈544/570 nm; used for AD drug screening and aggregation kinetics research
