Description RVG TFA is a 29-amino acid peptide fragment (residues 189-214) derived from the rabies virus glycoprotein (RVG), with the sequence YTIWMPENPRPGTPCDIFTNSRGKRASNG. It is provided as a trifluoroacetate (TFA) salt, and the free base has a CAS number of 115136-25-9.
RVG29 is one of the most widely studied blood-brain barrier (BBB) penetrating peptides. Its mechanism of action involves specific binding to the α7 nicotinic acetylcholine receptor (α7-nAChR), which is highly expressed on brain microvascular endothelial cells. This binding triggers receptor-mediated transcytosis (RMT), enabling efficient transport of conjugated drugs, nanoparticles, siRNA, or proteins across the blood-brain barrier into the central nervous system.
RVG29 contains a basic amino acid-rich region (Arg-Gly-Lys-Arg) that carries a positive charge at physiological pH, facilitating initial electrostatic interactions with negatively charged glycosaminoglycans on the cell membrane surface.
