Product name CAS 12705-41-8:Colistimethate sodium
Description Colistimethate sodium is a chemically modified prodrug of colistin (polymyxin E). Methanesulfonation neutralizes free amino groups, significantly reducing direct nephrotoxicity and neurotoxicity while greatly improving water solubility. It slowly hydrolyzes in vivo to release active colistin, which acts on Gram-negative bacterial outer membrane LPS and phospholipids, disrupts membrane integrity and leads to bacterial lysis. It exhibits potent bactericidal activity against multidrug-resistant Gram-negative bacteria such as Pseudomonas aeruginosa and Acinetobacter baumannii. Applied in drug-resistant Gram-negative infection mechanism research, antibiotic pharmacodynamic evaluation and antibacterial drug development.
Purity (HPLC, %) 90%/95%/98%
Average molecular weight (Da) ~1750 (multi-component mixture)
Structure type Cyclic lipopeptide prodrug, methanesulfonated modification, sodium salt form

Functional Description

Product Code Frontend Short Summary (for search list display) Detailed Biological Function Description Applicable Research Areas (comma-separated) Targets / Pathways
PEP-CMS001 Low-toxicity cyclic lipopeptide prodrug that activates in vivo to kill MDR Gram-negative bacteria, with significantly lower nephrotoxicity than colistin sulfate Colistimethate sodium is a classic polymyxin prodrug. By methanesulfonating free amino groups on colistin side chains, it shields the direct nephrotoxicity and neurotoxicity caused by positive charges while greatly improving water solubility. After entering the body, it slowly hydrolyzes in serum and tissues, gradually releasing biologically active free colistin. Active colistin binds to lipopolysaccharide (LPS) on the Gram-negative bacterial outer membrane via electrostatic interaction, displaces divalent cations on the membrane, disrupts outer membrane integrity, further penetrates to the inner membrane causing cytoplasmic membrane depolarization and cytoplasmic leakage, ultimately leading to bacterial death. This molecule exhibits potent bactericidal activity against clinical multidrug-resistant Gram-negative bacteria such as Pseudomonas aeruginosa, Acinetobacter baumannii and carbapenem-resistant Enterobacteriaceae, and is one of the core drugs for clinical pan-resistant Gram-negative infections.MDR bacterial infection research,Severe infection models,Gram-negative bacteria biology,Antibiotic prodrug development,Pseudomonas/Acinetobacter research,Antibacterial MDR bacterial infection research,Severe infection models,Gram-negative bacteria biology,Antibiotic prodrug development,Pseudomonas/Acinetobacter research,Antibacterial mechanism research Bacterial outer membrane lipopolysaccharide (LPS) / phospholipids; Outer membrane permeability disruption pathway; Bacterial plasma membrane depolarization lysis pathway
