Description

Shanghai Sonyt Biotechnology Co., Ltd. is one of the leading manufacturers and suppliers of fitc-β-ala-aβ (1-42), ammonium salt in China, also supports custom service. Welcome to wholesale bulk high quality fitc-β-ala-aβ (1-42), ammonium salt from our factory. Contact us for more details.

Description This is a FITC-β-Ala conjugate of the highly aggregation-prone, neurotoxic Aβ1-42; the flexible β-Ala spacer reduces fluorophore interference with self-assembly, and the ammonium salt aids aqueous reconstitution with low TFA carryover. It reports oligomerization, fibrillation and inhibitor/chaperone intervention in Alzheimer's in-vitro models.
Structure type Linear 42-mer (Aβ1-42, N-FITC-β-Ala, ammonium)
Solubility NaOH pre-dispersion then PBS pH adjust; protect from light
Storage Powder, −20°C, dry, light-protected

Physicochemical Solubility & Storage Parameters

Product Code Appearance Long-term Storage Short-term Storage Solution Stability Solid Powder Stability
PEP-FBAβ42-01 Yellow to tan powder (FITC-colored) Store at -20°C, sealed, desiccated, under argon/nitrogen, in single-use aliquots, ~2 years; PROTECT FROM LIGHT (amber vial / foil wrap); avoid repeated freeze-thaw. Abeta(1-42) is the most aggregation-prone Abeta isoform-prepare fresh, keep pH near neutral; avoid strongly acidic/basic conditions. 2-8°C, protected from light, for ~1 week; working aliquots at -20°C, light-protected, prepare fresh. Highly aggregation-prone and Met35 is oxidation-sensitive: pre-treat with a minimal volume of sterile DMSO or hexafluoroisopropanol (HFIP) to monomerize, then dilute into pH 7.4 PBS/HEPES to working concentration; handle in the dark. Use immediately after dilution for ThT-free fluorescence kinetics, TEM or cell-uptake assays. FITC is light-sensitive-minimize light exposure to prevent quenching. solid stable ~2 years at -20°C, light-protected, sealed, desiccated, flushed with Ar/N2; repeated freeze-thaw and light exposure accelerate aggregation, precipitation and fluorophore quenching.

Functional Description

Product Code Frontend Short Summary Detailed Biological Function Description Applicable Research Areas (comma-separated) Targets / Pathways
PEP-FBAβ42-01 FITC-labeled human Abeta(1-42) full-length fluorescent probe via a beta-Ala spacer, retaining the complete N-terminal hydrophilic region and strong hydrophobic C-terminus-used for Abeta aggregation kinetics, oligomer/fibril tracking, cell uptake and confocal imaging. Beta-amyloid (Abeta) is the principal component of senile plaques (SP) in Alzheimer's disease (AD) brain, generated by sequential cleavage of the amyloid precursor protein (APP) by beta-secretase (BACE1) and gamma-secretase. Abeta(1-42), bearing the strongly hydrophobic C-terminal Ile41-Ala42, aggregates and fibrillizes markedly faster than Abeta(1-40) and is the dominant isoform in toxic oligomers and senile plaques of AD brain. This reagent inserts a beta-Ala spacer at the N-terminus before FITC (fluorescein-5-isothiocyanate) coupling: the spacer reduces steric interference of the bulky dye with the beta-sheet nucleation core (central LVFFA residues 17-21 and the hydrophobic 30-42 transmembrane region), so the labeled peptide retains native aggregation behavior. FITC (Abs495/Em519) enables real-time monitoring of Abeta aggregation kinetics (intrinsic fluorescence or FRET with ThT), tracking of oligomeric/fibrillar species, and confocal/flow-cytometric studies of Abeta binding and internalization by neuronal membranes, microglia and receptors (RAGE, PrP(C), mGluR5, ApoE); it also supports high-throughput screening of anti-aggregation/depolymerization small molecules. Note: Abeta(1-42) is strongly amphipathic and aggregation-prone-store at -20°C in light-protected aliquots under inert gas, keep pH neutral, prepare fresh, and keep out of light to prevent FITC quenching. Alzheimer's disease (AD) mechanism, Abeta(1-42) aggregation/fibrillization kinetics, fluorescent tracking probe, Abeta oligomer/fibril imaging, cellular uptake/internalization, confocal/flow cytometry, anti-Abeta aggregation drug screening, FRET interaction studies Abeta aggregation core (LVFFA motif and C-terminal 30-42 hydrophobic region); RAGE, PrP(C), mGluR5, ApoE and other Abeta receptors/chaperones; FITC fluorophore (Ex495/Em519 nm)
