Description Biotin-KKKRKV acetate is the biotinylated acetate form of the classical SV40 large T-antigen NLS (KKKRKV). This basic hexapeptide is recognized by importin α/β to drive nuclear import; the biotin tag enables conjugation to nanoparticles, nucleic acids or protein cargoes for nuclear-targeted delivery and import-mechanism studies.
Peptide purity (HPLC) ≥95%
Molecular formula C58H113N21O16·xCH3COOH
Molecular weight (Da) ~1129 (free biotinylated peptide)
Structure  Linear 6-mer (KKKRKV NLS, biotinylated acetate)
Solubility Readily soluble in water/PBS
Storage Powder, −20°C, dry, light-protected

Physicochemical Solubility & Storage Parameters

Product Code Appearance Long-term Storage Short-term Storage Solution Stability Solid Powder Stability
PEP-BIKKKRKV-01 White to off-white powder Store at -20°C, sealed and desiccated, ~2 years. The strongly basic peptide is hygroscopic-keep tightly sealed and dry; store in a desiccator. 2-8°C for ~1 month; working aliquots at -20°C. Strongly basic peptide (4 Lys + 1 Arg) with excellent water solubility (mg/mL grade in pure water/aqueous buffer; no DMSO needed). Biotinylation retains high aqueous solubility; can be coupled to streptavidin-coated carriers/nanoparticles for nuclear-targeted delivery. No Cys/Met/Trp-chemically stable and resistant to oxidation. The acetate form is suitable for cell and in-vivo experiments. solid stable ~2 years at -20°C, sealed and desiccated; because of its strong basicity and hygroscopicity, weigh promptly after opening and reseal tightly; store in a desiccator.

Functional Description

Product Code Frontend Short Summary Detailed Biological Function Description Applicable Research Areas (comma-separated) Targets / Pathways
PEP-BIKKKRKV-01 N-terminally biotinylated SV40 large T-antigen canonical NLS hexapeptide KKKRKV, recognized by importin alpha/beta to drive nuclear import-used for nuclear-transport mechanism studies, cell-penetrating/nuclear-targeted delivery and streptavidin affinity purification. KKKRKV is the most classic and best-studied classical nuclear localization signal (NLS) from simian virus 40 (SV40) large T antigen. In the cytoplasm, NLS-bearing cargo is recognized by importin alpha (karyopherin alpha) via its ARM-repeat domain, then bound by importin beta to engage the nuclear pore complex (NPC); directional transport is driven by the Ran-GTPase gradient. As a minimal functional NLS, KKKRKV can be fused to proteins, peptides, nucleic acids, liposomes or nanoparticles to route exogenous cargo to the nucleus-it is the most widely used peptide signal in nuclear-targeted delivery. N-terminal biotinylation enables, via streptavidin bridging: (1) conjugation of the NLS to streptavidin-coated nanoparticles/liposomes/fluorescent microspheres to build nuclear-targeted delivery systems; (2) use as an affinity pull-down probe for importin alpha/beta nuclear transport receptors to study nuclear-import mechanisms; (3) as a cell-penetrating peptide (CPP) to assist macromolecular cellular uptake and nuclear entry. This strongly basic peptide (+5 side-chain charge) is highly water-soluble and supplied as the acetate salt, suitable for cell and in-vivo work; note that NLS function depends on importin alpha recognition-excess free NLS will competitively inhibit endogenous nuclear import. Nuclear transport (NLS) mechanism, importin alpha/beta nuclear import, SV40 large T antigen, nuclear-targeted delivery (drugs/genes/nanoparticles), cell-penetrating peptides (CPP), biotin-streptavidin affinity purification, macromolecular nuclear delivery Importin alpha (karyopherin alpha, the NLS receptor) / importin beta heterodimer nuclear transport receptors; nuclear pore complex (NPC); high-affinity binding to streptavidin
