CD31 is a type I transmembrane glycoprotein expressed on platelets, endothelial cells and leukocytes; its cytoplasmic tail bears two ITIMs that, upon TCR stimulation, become phosphorylated by Lck and recruit SHP-1/SHP-2 phosphatases to negatively regulate T-cell activation. TCR stimulation also induces juxtamembrane proteolytic cleavage (shedding) of CD31, releasing the ITIM-containing cytoplasmic domain.
